ABSTRACT

Objective:To observe the multimodal imaging characteristics of combined hamartoma of the retina and retinal pigment epithelium (CHRRPE).Methods:A cross-sectional study was conducted.Sixteen eyes of 16 patients with CHRRPE were enrolled in Second People's Hospital of Yunnan Province from March 2013 to July 2019.Fundus color photography, fundus autofluorescence (FAF), fluorescein fundus angiography (FFA), indocyanine green angiography (ICGA), optical coherence tomography (OCT), optical coherence tomography angiography (OCTA) and multicolor imaging were performed in all patients.The multimodal imaging characteristics were analyzed.This study protocol adhered to the Declaration of Helsinki and was approved by an Ethics Committee of Second People's Hospital of Yunnan Province (No.20130106). Written informed consent was obtained from patients or their guardians prior to any medical examination.Results:Tumors were located in the posterior optic disc, and translucent glial lesions with unclear borders and slight elevations were observed.The surface of the lesions was attached by different degrees of fibroproliferative membrane, and the adjacent vessels were twisted and dilated.The tumors presented flat bulging green reflexes on the retina at the posterior pole by multicolor imaging, and OCT image showed thickened optic disc and retina near the optic disc, structural disorder, high reflectance of the surface, and low reflectance of the deep retina below the periretinal membrane.OCTA showed irregular blood flow signals, and the signal of retinal blood vessels was twisted and dilated.FAF showed that the autofluorescence intensity of tumors was weakened to different degrees.Early lesions presented different degrees of blocked fluorescence in FFA.Deformed and tortuous blood vessels were found in the eyes, and telangiectasia showing needle-like punctate strong fluorescence leakage was observed in severe eyes by FFA.ICGA showed no abnormal choroidal vessels.Conclusions:The main imaging features of CHRRPE include abnormal retinal blood vessels in the tumor area and fibrous proliferative membranes on the surface in color image; OCT shows that the retina and the retinal pigment epithelium are involved, and the retina in the tumor is thickened with disordered structure; high reflection OCTA shows irregular internal blood flow signals inside the tumor; FFA examination shows fluorescence obscuration and obviously tortuous retinal blood vessels.Multimodal imaging examinations are helpful for the diagnosis of CHRRPE.

ABSTRACT

OBJECTIVE:To explo re the dose-effect relationship and mechanism of protective effects of total asiaticoside (TA) on gastrointestinal motility and enteric nervous system (ENS)in aged functional dyspepsia (FD)model rats. METHODS :Aged male SD rats of 16 months old were randomly divided into blank control group ,model group ,TA low dose ,medium dose and high dose groups (15,30,60 mg/kg),with 8 rats in each group. FD model was established by tail-stimulation combined with irregular diet for 4 weeks. The next day after modeling ,administration groups were given relevant doses of TA solution intragastrically ; control group and model group were given constant volume of normal saline intragastrically ,once a day ,for consecutive 15 d. Gastric emptying rate and small intestinal propulsion rate of rats were examined. ELISA were used to detect serum contents of MTL and VIP. Immunofluorescence and immunohistochemistry were proposed to measure the expression of ENS marker (S100β and GDNF)in gastric antrum tissue. The protein expression of S 100β,GFAP,PGP9.5,GDNF,p-MEK and p-ERK 1/2 in gastric antrum tissue were measured by Western blotting assay. RESULTS :Compared with blank control group ,gastric emptying rate and small intestinal propulsion rate ,serum MTL content and protein expression of PGP 9.5 in gastric antrum tissue of model and TA low,medium dose group were decreased significantly ,while serum VIP content ,protein expressions of S 100β,GFAP,GDNF, p-MEK and p-ERK 1/2 in gastric tissue were increased significantly (P<0.05). Compared with model group ,gastric emptying rate and small intestinal propulsion rate of TA groups were increased significantly (P<0.05);except for GFAP protein in TA low dose group(P>0.05),the serum MTL content and the expression of PGP 9.5 protein in gastric antrum tissue of rats in TA groups were increased significantly ,while serum VIP content ,protein expression of S 100β,GFAP,GDNF,p-MEK and p-ERK 1/2 in gastric antrum tissue were decreased significantly (P<0.05). Some or most of the content of gastrointestinal motility indexes and related factor protein expression were significantly different among TA groups (P<0.05),and the indexes in TA high dose group could recover to the levels which were not significantly different with blank control group (P>0.05). CONCLUSIONS :TA can dose-dependently improve the gastrointestinal motility deficiency and ENS dysfunction in aged FD model rats ,especially in high dose(60 mg/kg)of TA group. Its mechanism may be related with promoting the release of endogenous MTL ,inhibiting the secretion of VIP ,expression of GDNF and the activation of downstream signaling pathway ,and promoting the repair of ENS and intestinal neurons.

ABSTRACT

Objective: To investigate the effects of circulating exosomes derived from oral cancer patients on the proliferation, migration and invasion of tongue cancer CAL27 cells. Methods: Circulating exosomes were isolated from the peripheral blood of 20 oral cancer patients using gradient centrifugation. The characteristics of the exosome was studied by transmission electron microscope (TEM) and Western blot. After treatment with circulating exosomes, CAL27 cell proliferation was evaluated with MTT assay, migration was observed by cell wound scratch assay and the invasion of CAL27 cells was tested by Transwell assay. CAL27 cell bearing nude mice were used to estimate the effect of circulating exosome on CAL27 cell proliferation and metastasis in vivo. Results: The proliferation, migration and invasion of CAL27 cells were enhanced by circulating exosomes from oral cancer patients. In vivo study showed that the tumorigenesis and metastasis of CAL27 cells were enhanced by circulating exosomes. Conclusion: Circulating exosomes from oral cancer patients can promote the proliferation, migration, invasion and metastasis of CAL27 cells.

ABSTRACT

OBJECTIVE:To study the antioxidant effects of Scutellaria baicalensis(SBE)on the model mice with Alzheimer’s disease. METHODS:All mice were treated by D-gal (150 mg/kg) and NaNO2 (100 mg/kg) for 60 d to reproduce the AD model mice,ip,once a day. Totally 60 mice were randomly divided into normal control group(isovolumetric sodium chloride injection), model group (isovolumetric sodium chloride injection),estradiol (0.4 mg/kg) group,and SBE high,medium and low doses (500,250 and 125 mg/kg). The mice were intragastrically administrated while modeling except normal control group,once a day, for continuous 60 d. The superoxide dismutase(SOD),catalase(CAT)activities and malondialdehyde(MDA)content in brain tis-sue ,and glutathione peroxidase (GSH-Px) activity in serum of mice were determined. RESULTS:Compared with normal control group,the SOD,CAT activities in brain tissue,and GSH-Px activity in serum of mice in model group were decreased,MDA con-tent in brain tissue were increased,with significant difference(P<0.01 or P<0.05);compared with model group,the SOD,CAT activities in brain tissue and GSH-Px activity in serum of mice in SBE high and medium dose groups were increased,MDA content was decrease,with significant difference (P<0.01 or P<0.05). CONCLUSIONS:SBE can enhance the antioxidant capacity in AD model mice by a mechanism that may be related to the improvement of related oxidation indicators’levels.

ABSTRACT

Objective To evaluate local moderate hypothermia technology on acute brain hemorrhage cases.Methods 100 patients of cerebral hemorrhage were randomly divided into two groups(moderate hypothermia groupand control group). The moderate hypothermia group was 50 cases treated with moderate hypothermia after routine treatment,the control group was 50 cases with routine treatment. The two groups were assessed after treatment for 21 days. Results The group treated via moderate hypothermia therapy showed the nerve function damages relatively lower than the control group(P < 0.05), and the surviving probability and curative ratio were significant higher than the control group(P< 0.05 ). Conclusion The local moderate hypothermia therapy for acute cerebral hemorrhage patient in clinical can significantly improve the never cell function and reduce mortality and disablement.

ABSTRACT

Objectives: To study the inhibitory effects of grape juice on human prostate carcinoma PC-3 cells and its possible mechanism. Method: PC-3 cells were treated with grape juice in different concentration(16, 32, 64 ?l/ml) and resveratrol for 48 h; and the proliferation of PC -3 cells were measured by growth curve and MTT assay. TUNEL was used to observe the morphology of apoptotic cells and flow cytometry to analyze the PC-3 apoptosis. Results: Resveratrol and grape juice could markedly inhibit the proliferation of PC-3 cells, and TUNEL positive cells were detectable. The percentages of apoptotic cells were 6.0%,18.5%,30.0%, 12.7% in experimental groups and resveratrol group respectively. The effect on PC-3 cells was enhanced with increasing amount of grape juice. The effect of low dose group was weaker than that of resveratrol group, but medium and high dose groups were stronger. Conclusion: The anti-prostate cancer effect of grape juice is not only related to resveratrol contained but also to synergism with other components, and its mechanism might be the induction of PC-3 cells apoptosis.
